Uses
Mogrol is a biometabolite of mogrosides, and acts via inhibition of the ERK1/2 and STAT3 pathways, or reducing CREB activation and activating AMPK signaling.
Definition
ChEBI: Mogrol is a tetracyclic triterpenoid that is cucurbitadienol in which the side-chain double bond (position 24-25) has undergone formal oxidation to introduce hydroxy groups at positions 24 and 25 (the 24R stereoisomer). It is a biometabolite of mogrosides found in Siraitia grosvenorii. It has a role as an antineoplastic agent. It is a tetracyclic triterpenoid and a hydroxy seco-steroid. It is functionally related to a cucurbitadienol.
Biochem/physiol Actions
Mogrol is an aglycone of mogrosides from Siraitia grosvenorii that exhibits neuroprotective and anticancer activities. It induces apoptosis in K562 leukemia cells through suppression of ERK1/2 and STAT3 pathways. Mogrol appears to ameliorate the memory impairment caused by Aβ1-42 in mice. Mogrol increases AMPK activity in hepatocytes and suppresses adipocyte differentiation in 3T3-L1 cells.
Synthesis
1) Luo Han Guo dried fruit 100g, crushed into very coarse powder, add 600mL 70% industrial ethanol reflux extraction for 3h, filtration, filtrate recovery of ethanol (can be used again for the extraction of herbs), to get the alcohol extract 9.5g. . 2)Take the alcohol extract, add 100mL of water suspension, and then extract with 50mL water volume of water-saturated ethyl acetate, separate the ethyl acetate layer, decompression recovery of ethyl acetate (can be used again for extraction), get ethyl acetate extract 1.1g. . 3) The aqueous solution after ethyl acetate extraction was added to hydrochloric acid solution to make the concentration of hydrochloric acid up to 5%, and then 100mL of ethyl acetate was added and heated to reflux for 8h. Cooled to room temperature, the ethyl acetate solution was divided, and NAOH solution was added to the ethyl acetate solution, adjusting the pH to 7, and then separating the ethyl acetate layer, and recovering the ethyl acetate under reduced pressure (which can be used again and again) to get the extract containing rosmarinol 1.9g. . x000D_. 4).
