Paeoniflorin is an active compound of the Paeonia root. Paeoniflorin inhibits Aβ25-35-induced mitochondrial dysfunction, which includes decreased mitochondrial membrane potential, increased Bax/Bcl-2 ratio, cytochrome c release and activity of caspase-3 and caspase-9. Paeoniflorin could attenuate or restore the viability loss, apoptotic increase, and ROS production induced by Aβ25-35 in SH-SY5Y cells.

General Description

Paeoniflorin has been demonstrated to exert anti-inflammatory and immunomodulatory effects.

Biochem/physiol Actions

Herbal constituent of peony root. It is used in Chinese traditional medicine for its spasmolytic and pain-relieving properties. Exhibits anticoagulant, neuromuscular blocking, cognition-enhancing, immunoregulating, and antihyperglycemic effects.

Safety Profile

Moderately toxic by intravenous route. Mildly toxic by intraperitoneal route. When heated to decomposition it emits acrid smoke and irritating fumes.

Synthesis

Weigh 40kg white peony herbs, soaked in 70% ethanol overnight, reflux extraction 2 times, each time 2h, filtration, combined filtrate, rotary evaporation until no alcohol flavor, adjust the liquid so that each 1ml of liquid is equivalent to 0.5g of raw drug, standby. Weigh 120kg (wet weight) LX38 resin, loaded into a stainless steel column (300 2300cm), column volume of 60L, on the sample, adsorption of 1h, the first 300L of water, 10% ethanol 300L to remove impurities, and then 50% ethanol 360L elution of paeoniflorin and paeonia lactiflora paeoniflora glycosides, and finally 95% ethanol regeneration of resin, the flow rate of 150-200L/h, the 50% ethanol elution of peoniflorin and paeoniflorin. Finally, the resin was regenerated with 95% ethanol at a flow rate of 150-200L/h, and the 50% ethanol was concentrated and evaporated to obtain 2.52kg, in which the purity of paeoniflorin was 24% and that of paeoniflorin was 16%.
