Chemical Properties

The molecular formula of 2,8-DIAZA-SPIRO[4.5]DECANE-8-CARBOXYLIC ACID TERT-BUTYL ESTER is C₁₃H₂₄N₂O₂, with a molecular weight of 240.34 g/mol. It is a solid powder at room temperature, with a density of approximately 1.073 g/cm³, a boiling point of around 337.0°C, and a flash point of about 157.6°C. It is generally recommended to store it sealed, protected from light, at 2-8°C to maintain its stability.

Uses

tert-Butyl 2,8-Diazaspiro[4.5]decane-8-carboxylate is used in preparation of substituted aminoquinolones as DGKα inhibitors for immune activation.

Synthesis

The synthetic route for tert-butyl 2,8-diazaspiro[4.5]decane-8-carboxylate was as follows: the intermediate tert-butyl 4-(2,2-dimethyl-4,6-dicarbonyl-1,3-dioxin-5-ylidene)piperidine-1-carboxylate was subjected to Michell addition with nitromethane to give the compound tert-butyl 4-(2,2-dimethyl-4,6-dicarbonyl-1,3-dioxin-5-yl)-4-(nitromethyl)piperidine-1-carboxylate and then by catalytic hydrogenation and lactamization to give the compound 8-(tert-butoxy) -4-(carbonyl) piperidine 1-carboxylate. tert-butyl 4-(2,2-dimethyl-4,6-dicarbonyl-1,3-dioxane-5-yl)-4-(nitromethyl)piperidine-1-carboxylate was obtained by catalytic hydrogenation and lactamization, the compound 8-(tert-butyloxycarbonyl)-3-carbonyloxy-2,8-diazaspiro[4.5]decane-4-carboxylic acid was obtained by decarboxylation in the next step to obtain the product 3-carbonyloxy-2,8-diazaspiro[4.5]decane-8-carboxylic acid tert-butyl ester, and 4-(2-hydroxy)(2-hydroxyethyl)-4-(2,6-dicarbonyl)dioxine-8-carboxylic acid was prepared by ring opening reaction. The tert-butyl ester of 4-(2-hydroxyethyl)-4-(hydroxymethyl)-1-piperidinecarboxylate was prepared by ring-opening reaction, followed by the preparation of tert-butyl 4-(2-hydroxyethyl)-4-(hydroxymethyl)-1-piperidinecarboxylate 1,1-dimethylethyl ester, and then tert-butyl 2,8-diazaspiro[4.5]decane-8-carboxylate was prepared by intramolecular dehydration.
