Chemical Properties

Hemihydrate: white foamy substance. [α]D26-29.5° (C=0.2, ethanol). Spirapril Hydrochloride (Spirapril Hydrochloride): C22 H3ON2O5S2?HCl.[94841-17-5]. White solid, melting point 192-194°C (decomposition). [α]D26-11.2° (C=0.4, ethanol). Spiraprilat (Spiraprilat): C20H26N2O5S2.[83602-05-5]. Diacid for spirapril. Semihydrate, melting point 163～165°C. [α]D26+4.1° (C=0.4, ethanol).

Uses

Spirapril is an Angiotensin-converting enzyme inhibitor and used to treat high blood pressure.

Definition

ChEBI: Spirapril is a dipeptide, a dithioketal, an azaspiro compound, a dicarboxylic acid monoester, an ethyl ester, a tertiary carboxamide, a secondary amino compound and a pyrrolidinecarboxylic acid. It has a role as a prodrug, an EC 3.4.15.1 (peptidyl-dipeptidase A) inhibitor and an antihypertensive agent. It is functionally related to a spiraprilat.

in vivo

Spirapril (feeding needle; 10 mg/kg; 3 weeks) decreases alcohol intake in TGM123 mice and dose not reduce the alcohol consumption in TLM mice[2].
Spirapril shows a 40.2% reduction in ACE activity in brain membrane from treated-mice[2].
Spirapril can crosses the blood-brain barrier and suppresses the transgene effect in the experiments[2].
Spirapril prevents left ventricular hypertrophy, decreases myocardial damage and promotes angiogenesis in spontaneously hypertensive rats[3].
