Uses

5-Acetamido-2-chlorobenzoic Acid is used for preparation of 2-amino-5-substituted pyrimidine kinase inhibitors and use in therapy.

Synthesis

1. In a dry reaction flask, 5-amino-2-chlorobenzoic acid (0.50 g, 2.9 mmol) was dissolved in dichloromethane (15 mL). 2. Acetyl chloride (0.30 mL, 4.2 mmol) and triethylamine (1.2 mL, 8.6 mmol) were added sequentially to the above solution. 3. The reaction mixture was stirred at room temperature for 2 h. When the reaction was complete, the mixture was slowly poured into water. 4. The aqueous phase was extracted with ethyl acetate and the organic layers were combined. 5. The combined organic layers were washed sequentially with water and saturated saline. 6. 6. The organic layer was dried over anhydrous sodium sulfate and filtered to remove the desiccant. 7. The filtrate was concentrated under pressure to give 5-(acetylamino)-2-chlorobenzoic acid (0.17 g, 27% yield) as a brown solid.
