Lithium dodecyl sulfate Usage And Synthesis
Description
Lithium dodecyl sulfate (LDS) is an anionic detergent and surfactant that is frequently used in electrophoresis and chromatography. LDS may be substituted for sodium dodecyl sulfate (SDS) in electrophoresis under cold conditions. The use of LDS in micellar electrokinetic chromatography has been extensively studied. A report has been published on the partition equilibria of LDS between water and bilayer membranes. LDS has been utilized in protein electrophoresis and proteomics studies. The three-dimensional structure of the amyloid β-protein fragment 25-35 has been investigated in LDS micelles by two-dimensional 1HNMR. LDS has been used to induce changes in protein conformation.
Chemical Properties
Lithium dodecyl sulfate is a white, shiny crystalline powder or flake, soluble in organic solvents such as alcohols, ethers, and hydrocarbons, but only slightly soluble in water. It exhibits good thermal stability and remains stable at room temperature. As an anionic surfactant, it demonstrates excellent surface activity.
Uses
Lithium dodecyl sulfate has been used as a component of lysis buffer.
Uses
Anionic detergent that may be used in place of SDS for electrophoresis in cold conditions.

Purification Methods
Recrystallise this detergent twice from absolute EtOH and dry it under vacuum. Critical Micellar Concentration (CMC) in H2O is 8.77 x 10-3M. [Mukerjee et al. J Phys Chem 71 4166 1967, Mysels & Dulin J Colloid Sci 10 461 1955, Beilstein 1 IV 1847.]
Precautions
In general, autoclaved lauryl sulfate solutions undergo hydrolysis to yield lauryl alcohol, which precipitates from solution, and sulfuric acid, which decreases the solution pH. It is not advised to autoclave lauryl sulfate solutions.
