PACAP 1-27 Usage And Synthesis
Chemical Properties
This product is made from the posterior lobe of the pituitary gland of pigs, cows, sheep and other animals by dehydration, drying and fine grinding. It contains oxytocin (oxytocin, oxytocin, about 20 units) and a small amount of vasopressin (vasopressin, 0.5 units), both of which are very similar in structure, and both are natriuretic peptides containing disulfide bonds. The powder is a pale yellow or pale gray non-crystalline powder with a peculiar odor and is practically insoluble in water. It has the effect of excitation of the uterus and promotion of milk discharge, and the vasodilatation of blood vessels when the dose of contraction is large, but the vasoconstriction of blood vessels when the dose of ascending hormone is large, as well as the diuretic effect.
Uses
Pituitary Adenylate Cyclase-Activating Polypeptide 1-27 (PACAP 1-27) is a neuropeptide that stimulates adenylyl cyclase.
Biological Functions
PACAP 1-27 is an endogenous neuropeptide and is the C-terminally truncated form of PACAP 38. PACAP 27 has considerable homology with vasoactive intestinal peptide (VIP) but is >100 fold more potent than VIP as an agonist of the PAC1 receptor. PACAP 27 functions in the control of anterior pituitary hormone secretion, vasodilation, adrenaline secretion, insulin secretion and immunosuppression.
Biological Activity
PACAP 1-27, also known as PACAP 27 Amide, is an endogenous neuropeptide showing considerable homology with vasoactive intestinal peptide (VIP). Potently stimulates adenylyl cyclase.
in vivo
The inhibitory effect of pituitary adenylate cyclase activating polypeptide (PACAP (1-27), human, ovine, rat) on the increase in total pulmonary resistance (RL) causes either by allergen or histamine in anaesthetized, ventilated guinea-pigs is studied. PACAP (1-27), human, ovine, rat given via i.v. infusion (0.045-4.5 nmol/kg/min) dose-dependently reduces the increase in RL caused by inhaled ovalbumin and histamine. At the highest dose, PACAP (1-27), human, ovine, rat prevented the increase in RL caused by ovalbumin and histamine completely. Infusion of PACAP (1-27), human, ovine, rat and the β2-adrenoceptor agonist, salbutamol (0.045-4.5 nmol/kg/min) inhibit the increase in RL similarly, but salbutamol increases the heart rate more than PACAP (1-27), human, ovine, rat[3].
