General procedure for the synthesis of 5-amino-6-chloronicotinic acid methyl ester from 6-chloro-5-nitronicotinic acid: a suspension of methyl 6-chloro-5-nitronicotinic acid (1 eq.), iron powder (5.2 eq.) and NH4Cl (5.3 eq.) in methanol was heated and reacted for 2 hr at 75 °C. Upon completion of the reaction, the mixture was filtered through a diatomaceous earth pad while hot and the filtrate was subsequently concentrated in vacuum to afford the target product methyl 5-amino-6-chloronicotinate in 56% yield.
