Uses
1-(2,3-Dimethylphenyl)ethyl Chloride is an impurity of Medetomidine (M203250), an a2-Adrenergic agonist. Sedative
Synthesis
In a 1000mL three-necked flask, add tetrahydrofuran (THF) 200mL and magnesium strips 12g (0.5mo1), heat and reflux, dropwise add 2,3-dimethylbromobenzene 92.5g (0.5mo1) of THF solution 150mL, reflux after 1h; cooled to room temperature, dropwise add acetaldehyde 36mL (0.64mol)/THF 100mL, continue to reflux 1h , evaporate THF under reduced pressure, cooled and slowly added NH4Cl aqueous solution (25g NH4C1 + 80mL water) and ethyl acetate 300mL. static layering, dry the organic phase with anhydrous Na2S04, filtration, evaporation of solvents, and then distillation under reduced pressure to collect the fraction of 115-118 ??/300Pa. Put anhydrous ZnCl2200g and concentrated HCl300mL, fully dissolved and added the above fractions 150g (1mo1), stirred vigorously for 4h. Layering, anhydrous CaC12 to dry the organic phase, filtered, filtrate evaporated to remove the solvent, and then distilled under reduced pressure to collect 80 ~ 83 ?? C / 300Pa fractions to obtain the product 1-(1-chloroethyl)-2,3-dimethylbenzene (142g. Yield 84.2%).
