Step 1: Synthesis of 5-bromo-1-methylpyrimidin-2(1H)-one A mixture of 5-bromopyrimidin-2(1H)-one (3 g), iodomethane (1.1 mL) and potassium carbonate (2.4 g) in N,N-dimethylformamide (60 mL) was stirred for 5 hours at room temperature. After completion of the reaction, the mixture was filtered and the filtrate was concentrated. The residue was partitioned between water and dichloromethane. The organic phase was washed with brine, dried over anhydrous magnesium sulfate, and then concentrated to afford the target product 5-bromo-1-methylpyrimidin-2(1H)-one. Yield: 385 mg; liquid chromatography (Method 9): retention time t R = 0.25 min; mass spectrometry (ESI+): m/z = 189 [M + H]+.
