Synthesis of 6-(benzyloxy)-7-methoxyquinazolin-4(3H)-one: 2-amino-5-(benzyloxy)-4-methoxybenzoic acid (5 g, 18.3 mmol) was mixed with formamidine acetate (3.8 g, 36.6 mmol) in ethylene glycol monomethyl ether (25 mL). The reaction mixture was heated at 130°C for 24 hours. After completion of the reaction, it was cooled to room temperature and concentrated in vacuum to remove some of the solvent. Subsequently, ammonium hydroxide (5 mL, 30% aqueous solution) and water (50 mL) were added. The precipitated solid was collected by filtration, washed with water and hexane, and dried under vacuum to give 4.87 g of 6-benzyloxy-7-methoxyquinazolin-4-one in 94% yield. Mass spectrum (AP/CI): 283.1 (M+H)+.
