Description

Propargyl-PEG5-alcohol enables Click Chemistry reactions with azide compounds or biomolecules via copper catalyzed azide-alkyne Click Chemistry to yield a stable triazole linkage.

Uses

Propargyl-PEG4-alcohol is a PEG-based PROTAC linker can be used in the synthesis of PROTACs[1]. Propargyl-PEG4-alcohol is a click chemistry reagent, it contains an Alkyne group and can undergo copper-catalyzed azide-alkyne cycloaddition (CuAAc) with molecules containing Azide groups.

Synthesis

In a three-necked flask equipped with a magnetic stirrer, a reflux condenser tube and a thermometer, 1.4 g of potassium tert-butoxide, 2.0 g of tetrapolyethylene glycol, 3.0 g of propargyl bromide and 95 mL of tetrahydrofuran were added respectively, protected by nitrogen, heated with a water bath at a reaction temperature of 30 ??, and the reaction was carried out for 24 h. The product was extracted with saturated aqueous saline solution, and the supernatant layer of the liquid was evaporated by rotary evaporation, and dried in a vacuum to obtain the viscous liquid product with a yield of 86.1%. The obtained product was subjected to a curing reaction with a certain azide curing agent with a functional degree of 3.82. Using CuI as a catalyst and triglyceride acetate as a plasticizer, the cured product, propargyl-tetramethylene glycol, was obtained.
