Uses
Dibenzyl phosphate (DBzP) can be used:
- To promote the monoselective ortho-C-H alkylation of N-quinolyl benzamides with primary and secondary alkyl iodides.
- For the ring-opening reaction of epoxide such as benzylglycidol to synthesize dihydroxyacetone phosphate (DHAP).
- As a reactant for the synthesis of stereospecific 1,2-trans glycosyl phosphates.
Definition
ChEBI: Dibenzyl phosphate is a dialkyl phosphate.
Application
Dibenzyl phosphate and its derivatives are phosphorylation reagents; for example, di-p-tolyl phosphate is one of the key intermediates in the synthesis of fosapitant dimethylglucamine.
Synthesis
Dibenzyl phosphate with an overall yield of 67.4% was synthesized via a three-step reaction using benzyl alcohol and phosphorus trichloride as starting materials.
(1) Benzyl alcohol, phosphorus trichloride, and triethylamine were reacted in toluene to form dibenzyl phosphite (purity: 69%);
(2) Dibenzyl phosphite and aqueous sodium hydroxide were reacted in carbon tetrachloride to form dibenzyl phosphate sodium salt tetrahydrate (purity: 95%), with a two-step yield of 46.8%;
(3) Dibenzyl phosphate sodium salt tetrahydrate was reacted with hydrochloric acid to form dibenzyl phosphate (purity: 97%), yield: 85.6%;
(4) The product was refined with ethyl acetate/petroleum ether = 1:3 (purity: 99.5%), yield: 88.1%.
Precautions
dibenzyl phosphate may be stored at 2–8 °C. It is incompatible with strong oxidizing agents. Avoid contact with skin and eyes.
