Uses

3,3′-Diaminobenzidine (DAB) Enhanced Liquid Substrate System tetrahydrochloride has been used:

in the detection of hydrogen peroxide in rice leaves
as a substrate for rabbit peroxidase in immunohistochemistry of mice brain sections
in immunocytochemistry of muscle-derived stem cells (MDSCs)

Uses

3,3′-Diaminobenzidine tetrahydrochloride has been used for immunohistochemistry.

General Description

3,3′-Diaminobenzidine (DAB) is a chromogenic substrate for peroxidase enzyme. It is converted to an insoluble precipitate after oxidation by peroxidase.

Biochem/physiol Actions

3,3′-Diaminobenzidine (DAB) is a chromogenic substrate for peroxidase enzyme. It is converted to an insoluble precipitate after oxidation by peroxidase. This feature is exploited for visualization in histochemical (HC) and immunohistochemical (IHC) staining.

Safety Profile

Suspected carcinogen withexperimental neoplastigenic and tumorigenic data. Poisonby intraperitoneal route. When heated to decomposition itemits very toxic fumes of HCl and NOx.

Purification Methods

Dissolve the salt in water and precipitate it by adding conc HCl, then drying it over solid NaOH. [Beilstein 13 IV 530.]
