Chemical Properties
Dark red, crystalline powder or dark red crystals, very hygroscopic.
Uses
Physiological analog of vitamin B12 where the CN group is replaced with OH. Exists in aqueous solution as an equilibrium mixture of the hydroxy isomer and the ionic aqua isomer (aquacobalamin). Precursor of the coenzymes methylcobalamin and cobamamide. Coordination Compound. Vitamin (hematopoietic).
General Description
Hydroxocobalamin is an analog of vitamin B12 with OH as an alkyl group. Vitamin B12 cobalamin refers to a group of chemically-related cobalt containing molecules.
Biochem/physiol Actions
Vitamin B12 cobalamin is involved in DNA synthesis and fatty acid synthesis. It also plays a vital role as a coenzyme in the conversion of mitochondrial methylmalonyl co-enzyme A to succinyl co-enzyme A. Vitamin B12 cobalamin is also involved in neurotransmitter synthesis and erythropoiesis. It also functions in the synthesis and maintenance of myelin sheath. Hydroxocobalamin (vitamin B12a, OHCbl) and cyanocobalamin (CNCbl) are storage and delivery forms.
Synthesis
2.4Kg methylcobalamin was prepared into 2000μg/ml methylcobalamin solution, PH was adjusted to 3.18 with 20% acetic acid, air stirring was turned on, and the content of hydroxocobalamin acetate in the reaction solution was 98.3% under the illumination of 17,000LX, and the reaction solution was adsorbed on the column of styrene large-pore adsorbent resin at the adsorption rate of 100L/h, and the layer was spread at the rate of 100L/h after washing with 450L acetone solution (in which acetone??water=5??95). The reaction solution was adsorbed on styrene macroporous adsorption resin column, the adsorption speed was 100L/h, after water washing, 450L of acetone aqueous solution (acetone:water=5:95) was used to spread the layer, and the spreading speed was 100L/h. When the color of the outlet was changed from light red to colorless, 350L of 40% of the acetone aqueous solution was resolved through the resin column at a speed of 100-250L/h, and the red crystalline solution was collected, and mixing was turned on, and 600L of acetone with the density of 0.810g/ml was added to the crystalline solution. Then add 600L of acetone solution with a density of 0.810g/ml into the crystallized stock solution, stir for 1h, leave it for 4h, and get 2.39Kg of hydroxocobalamin acetate after filtration, the product yield is 99.6%, the content is 98.22%, and the related substance is 1.20%.
