Uses

Ophiopogonin D is a natural product isolated from Radix Ophiopogon japonicus that protects the heart against doxorubicin-induced autophagic injury by reduction of both ROS generation and the disruption of the mitochondrial membrane damage.

Application

Ophiopogonin D is a bioactive compound that is found in Salvia miltiorrhiza. It has been shown to have hypoglycemic effects and significant cytotoxicity against tumor cells, which may be due to its ability to inhibit glucose uptake. It also causes mitochondrial membrane potential reduction, leading to the induction of apoptosis. This compound also has immunomodulatory effects. In addition, it inhibits the activation of toll-like receptors and prevents the translocation of nuclear factor-κB into the nucleus. Ophiopogonin D also has an effect on bowel disease, myocardial infarcts, and energy metabolism by inhibiting c-jun phosphorylation.

Biological Activity

Ophiopogonin D is a steroidal glycoside extracted from Ophiopogon japonicas th at promotes antioxidative protection of the cardiovascular system. Ophiopogonin D protects the heart against doxorubicin-induced autophagic injury by reduction of both ROS generation and the disruption of the mitochondrial membrane damage.

Synthesis

Take maitake dried herbs 500g, crushed through a 40-mesh sieve, add 4L 80% ethanol solution soaked for 0.5 hours, and then placed in a microwave device irradiation extraction for 15min, extraction temperature of 60 ~ 70 , microwave device power set to 200W, the extract was concentrated to get the extract of 68g, add 150ml of water dispersed, and then add 200ml of water saturated with n-butanol extraction for 2 times, and merged. The extract was extracted twice with 200ml of water saturated with n-butanol, combined with 300ml of acetone precipitation, filtered, and the filtrate was evaporated to obtain the total saponin of Ophiopogon 11.8g.

The chloroform, methanol, and 1% ammonia were measured at the ratio of 7:13:5 by volume, and then added into the dispensing funnel after ultrasonication and mixing, and then the upper and lower phases were separated by static, and then the upper phase was taken as the stationary phase, and the lower phase was taken as the mobile phase, and then the yellow powder was dissolved with 500ml of the lower phase and then pumped to the chromatographic column at a rate of 2.5~2.0 m. Chromatographic column, and then pumped into the mobile phase at a flow rate of 2.0 ml/min, after equilibrium, set the chromatograph speed of 850 r/min, injection through the injection valve, each injection volume of 150 mg, collect the effluent, evaporation under reduced pressure that is, maitake saponin D2.7g, content 99.3%.
