Definition
ChEBI: A methylguanine in which the methyl group is positioned on the oxygen at position 6. Formed in DNA by alkylation of the oxygen atom of guanine, most often by N-nitroso compounds and sometimes due to methylation by other compounds such as endogenous S-adenosylmethionine, it base-pairs to thymine rather than cytidine, causing a G:C to A:T transition in DNA.
Application
6-O-Methyl-guanine is a key molecular tool for studying DNA damage, repair mechanisms, and cancer chemotherapy. It is a potent mutagenic and carcinogenic DNA damage product induced by alkylating agents (such as certain environmental carcinogens and chemotherapeutic drugs). It can mispair with thymine (T), leading to G:C to A:T base transition mutations during DNA replication. Therefore, O⁶-MeG is widely used in in vitro and cellular experiments to simulate DNA damage states and study how cells remove this damage through repair proteins such as O⁶-methylguanine-DNA methyltransferase (MGMT).
